Using this guide https://www.clearos.com/resources/documentation/clearos/content:en_us:kb_o_adding_send_as_functionality_in_zarafa
I created the ldif file and used the Windows ldapadmin utility to import the file, which seems to have taken:
# zarafa-admin --list-sendas user1
Send-as list (1) for user user1:
Username Fullname
-------------------------------
user2 User Two
Message header as sent shows "user2@domain.com on behalf of user1@domain.com"
Restart zarafa and also tried a reboot.
What have I missed?

So I shut the VM down last night and fired it up again just now and the messages that I sent yesterday that were sitting in the Inbox with "on behalf of" have magically changed to the "sendas" originator.
So just to make sure I'm not going completely mad I did a new "sendas" message and it now works as intended. It seems that a reboot didn't make it work, but a complete shut down did???
